Summary:
The BCA method is a protein quantification technique that leverages a two-step chemical process. First, in an alkaline medium, proteins reduce cupric ions () to cuprous ions () through the biuret reaction. This reduction is primarily facilitated by peptide bonds and the side chains of specific amino acids like cysteine, tryptophan, and tyrosine. Subsequently, the sodium salt of bicinchoninic acid chelates with the newly formed ions, producing a purple-blue color. The intensity of this color is directly proportional to the protein concentration and is quantified by measuring the absorbance at 562 nm.
